Lethal Concentration of Ammonia and Tolerable Range of pH in zebrafish (Danio rerio)

Authors

Abstract:

Increasing in water ammonia is one of the major problems in aquaculture. Nitrogen-containing materials are producing in different ways, including oxidation of nitrogen-containing organic matters such as proteins, manure decomposition and ammonia excretion by fish. The increase in these compounds in water recirculation systems and aquaria has always been considered as a stress-causing factor. On the other hand, either increasing and/or decreasing in ::union::ized ammonia are affected by pH changes. In order to obtain the LC50 of ammonia, zebrafish were exposed to seven treatments including control, 0.069, 0.138, 0.276, 0.552, 1.105 and 2.210 mg/L of ::union::ized ammonia. Then, in order to obtain a tolerable pH range fish were exposed to either gradual increase or decrease of pH (0.1 units per minute) using HCl and NaOH 0.4 N. The amount of LC50 for different time periods was determined using the probit software. The amounts of LC50 for 24, 48, 72 and 96 hours were 0.766, 0.634, 0.532 and 0.471 mg/L, respectively. Maximum and minimum average of pH tolerance threshold was 11.64±0.04 and 2.60±0.09 respectively. The results showed that zebrafish is a sensitive species to ammonia poisoning, but it is resistant to pH changes.

Histopathological Changes of Zebrafish (Danio rerio) Ovaries Exposed to Sub-lethal Concentrations of Methyl Paraben

Paraben compounds are used widely as preservatives to inhibit microbial growth and extend used in pharmaceuticals, hygienic, cosmetics, care products. They pose the potential for reproductive toxicity for users. This paper provides an overview of the applications of histopathological biomarkers in reproductive toxicity in zebrafish caused by methylparaben. The present study investigated the cha...

Ammonia excretion by the skin of zebrafish (Danio rerio) larvae.

The mechanism of ammonia excretion in freshwater teleosts is not well understood. In this study, scanning ion-selective electrode technique was applied to measure H(+) and NH(4)(+) fluxes in specific cells on the skin of zebrafish larvae. NH(4)(+) extrusion was relatively high in H(+) pump-rich cells, which were identified as the H(+)-secreting ionocyte in zebrafish. Minor NH(4)(+) extrusion wa...

Danio Rerio, the Zebrafish

The zebrafish has long been a favourite model for the study of vertebrate development. Here we provide an overview of the current state of knowledge and resources for the study of this fish, with comments on the future direction of zebrafish genomics from Professor Mark Fishman and Dr Stephen Wilson.

BIOLOGY OF ZEBRAFISH, Danio rerio

Zebrafish, Danio rerio (Hamilton), is one of the most important vertebrate model organism in genetics, developmental biology, neurophysiology and biomedicine (Rubinstein, 2003; Amsterdam and Hopkins, 2006). It is a small, robust fish, so large numbers can be kept easily and cheaply in the laboratory, where it breeds all year round. Development is rapid, with precursors to all major organs devel...

Determining the lethal concentration of formalin to zebra fish (Danio rerio)

Formalin is an effective solution for surface and equipment disinfection and also for several aquaculture industry challenge. Since various aquatics have different sensitivity to this substance, determination of a precise concentration for cultured fish is important. Because of diversity in aquarium fish species, this topic more highlights. In this study, the method of The Organisation for Econ...

Pharmacological analysis of zebrafish (Danio rerio) scototaxis

The scototaxis test has been introduced recently to assess anxiety-like phenotypes in fish, including zebrafish. Parametric analyses suggest that scototaxis represents an approach-avoidance conflict, which hints at anxiety. In this model, white avoidance represents anxiety-like behavior, while the number of shuttling events represents activity. Acute or chronic fluoxetine, buspirone, benzodiaze...
